He's also quite obviously laughing internally at Midnighter's exchange with the horse. It's only politeness that keeps him from laughing outright.

"If you take the cart, I'll manage the horse."

As a samurai, he's an experienced equestrian. Just to be safe, Takatora approaches the horse from the front when he moves in to grab hold of its reins.
